(Fiona and Kento are coming home from school. On the way, Kento noticed a sign he had never seen before.)

Not again!
「また~!」というニュアンスで使う表現です。
気の進まないことや、いやなことが再び起こったときに使います。
(It is used when something you don’t like or something unpleasant happens again.)
たとえば、きのう友だちとクイズをしていたのに、
We have a quiz today. と言われたときに、
Not again! と答える感じです。
